Biotechnology for isolating lysozyme from bird egg white

Purpose. To create new drugs with antibacterial, antifungal, anti-inflammatory and immunomodulatory effects. For the isolation and purification of c- and g-type lysozyme from bird egg whites; purification of enzymes using gel chromatography. Determination of the protein fractional composition, molecular weight of isolated and purified drugs using PAGE and MALDI electrophoresis; study of spectral characteristics (UV spectroscopy, luminescence).

Application area. Protein chemistry, enzymology, molecular biology, biotechnology, pharmacy for use in medical practice.

Advantages. There are no analogues in Ukraine.

Description. New approaches to the production of c- and g-type lysozymes in laboratory conditions have been proposed. A modification of the laboratory method for isolating c-type lysozyme from chicken and quail egg whites has been proposed, which makes it possible to obtain an enzyme with a purity of 98% and a hydrolytic activity of 22025 units/mg and corresponds to the branded drug “AppliChem”. In order to develop a method for isolating g-type lysozyme, the influence of volumetric ratios of protein: solvent and temperature conditions of the process was studied. It was revealed that for g-type lysozyme, the use of the method of thermal denaturation of impurity proteins is inappropriate due to the loss of activity. Using gel chromatography with a preliminary lyophilization step, partially purified g-type lysozyme with a hydrolytic activity of 1333 U/cm3 was obtained.
