MULTI-BIO-SENSOR DEVICE BASED ON THE ARRAY OF ION-SENSITIVE FIELD TRANSISTORS “ISFET-2”

PURPOSE:

The Ukrainian Institute has developed a multi-biosensor device designed to determine the concentrations of toxic substances – pesticides and heavy metals in aqueous solutions.

DESCRIPTION:

The device relates to the field of ecology and can be used, in particular, for express analysis of wastewater, including near places of possible pollution, for assessing the presence and determination of the concentration of heavy metal ions, carbamate herbicides and organophosphate pesticides, and more specifically, for the design analytical systems for monitoring toxic substances in aqueous solutions. Application areas of the device: environmental monitoring: determination of general toxicity of solutions and simultaneous selective determination of some toxic substances, such as formaldehyde, hypochlorite, organophosphoric pesticides, carbamate herbicides, heavy metal ions; food quality control: determination of natural harmful substances (steroid glycoalkaloids) in food (vegetables, fruits) diagnostics in medicine: express determination of the main human metabolites (glucose, urea, creatinine) in biological fluids (blood, serum, blood plasma, urine) … The device contains five sensitive elements. Each sensing element contains a pH-sensitive field-effect transistor with a working membrane and a pH-sensitive field-effect transistor with a control membrane. The working membrane of the sensing element contains one of the enzymes that are sensitive to toxic substances. Acetylcholinesterase, butylcholinesterase, urease, glucose oxidase and mutarotase-invertase-glucose oxidase are used as such enzymes. The control membrane contains bovine serum albumin. The outputs of the sensor’s sensitive elements are connected to the inputs of the device for potentiometric analysis.

ADVANTAGES:

The proposed multi-biosensor uses a matrix of pH-sensitive field-effect transistors; there is a possibility of using a differential measurement method and using a larger number of different enzyme systems for selective assessment of the content of heavy metals and pesticides in the samples under study. Due to this, the device has a higher sensitivity and selectivity in comparison with analogues and allows obtaining results with a higher degree of reliability.

TECHNICAL AND ECONOMIC EFFECT:

The number of channels: 12 (2×6). The working volume of the flow system was 200 μL. Single measurement time: 0.1 sec. Operating temperature range: 0-40 ° С Time required for biochemical analysis: no more than 10 minutes. Automatic supply of analytes at a rate of 5 μl – 200 ml/min. Serial interface: RS232. Software: Win 9X, DOS. Power supply: 12V / 0.2A.

RESULTS OF THE RESEARCH:

Compared to analogs, the device has a higher sensitivity and selectivity and makes it possible to conduct a selective quantitative analysis of toxic substances in real samples.
